The Cytek Aurora CS system provides remarkable flexibility, enabling the use of a wide array of new fluorochrome combinations without requiring the reconfiguration of the detectors. Sharing the same optical system as the Cytek Aurora analyzer allows for the easy transfer of experiments from the analyzer to the Aurora CS system without the need to modify your panels. The state-of-the-art optics and low-noise electronics ensure high resolution and high sensitivity for identifying and isolating rare cell populations deep into the gating hierarchy.

The result is a system that delivers exceptional performance at the single-cell level which can resolve the most challenging cell populations, such as cells with high autofluorescence or low expression of key biomarkers, regardless of assay complexity. Equipped with advanced sorting features and monitoring tools, the Cytek Aurora CS system can isolate viable and functional cells for downstream studies.

Cytek's 40-color deep immunophenotyping panel described in the OMIP-069 publication (PMID: 32830910) can perform equally well on the Cytek Aurora CS system. As shown, a UMAP scatter plot from each system is overlaid, demonstrating a high level of comparability. Data from the Cytek Aurora analyzer (blue) and the Cytek Aurora CS system (orange), exhibit the same population patterns and overlap. In-Depth Sorting Functionality

The Cytek Aurora CS system combines FSP technology and high-end sorting functionality to dig deep into cellular assays. Sort populations up to 64 levels deep into the gating hierarchy, up to two-way sorting with 15 mL tubes, sort up to 6 ways into 1.5 mL or 5 mL tubes, or sort into 384-well or 96-well plates with index sorting capability. Live view camera monitoring helps monitor side-stream position, and other built-in automated tools ensure successful sorting.

In-Depth Purity Sorting

To demonstrate the advanced sorting capability of the Cytek Aurora CS system, a 28-color human deep T-cell and NK cell immunophenotyping panel was optimized and transferred from the Cytek Aurora analyzer. Six low frequency T-cell and NK cell subsets shown below were sorted with a 100 μm nozzle and then evaluated for purity. Download the brochure to see the sort results.

Functional Assays

When sorting cells for downstream experimental assays, it is crucial that the sorted cells remain viable and functional after sorting. The Cytek Aurora CS system is designed to sort cells as quickly and efficiently as possible while maintaining good cell viability and functionality after sorting. Human PBMC samples were prepared and sorted 4-ways with a 70 μm nozzle to collect CD4+ effector memory, CD8+ effector memory, TCRγδ T cells, and NK cells using the gating strategy below. Sort gates are outlined in orange.

After sorting, the cells were rested for 2 hours in an incubator, stimulated with PMA and ionomycin in the presence of BFA, monensin, and CD107a, then stained with 8 different intracellular markers. Download the brochure to see the results.

Autofluorescence Extraction

The Full Spectrum Profiling technology employed by the Aurora CS system can improve data clarity and resolution with autofluorescence extraction. Certain sample types, such as yeast and tumor samples, present difficulty in resolving due to their highly autofluorescent nature. For applications involving highly autofluorescent particles, the SpectroFlo® CS software autofluorescence extraction tool achieves new levels of resolution and increases sorting performance of challenging samples.
